Customer interview at Prince of Songkla University as to the benefits of the Smart Evaporator C10

Location of the unit :

Department of Pharmacognosy and Pharmaceutical Botany,
Prince of Songkla University (Thailand)

Interviewed with :

Ms. Similan Inaugsorn

-Master degree student , Department of Pharmacognosy and Pharmaceutical Botany.

              Name/Quantity of Solvent : 85% Ethanol

Type of container : 2 ML vial HPLC

              Temperature setting : 70°C

              Operation hour : 10-15 min

Mr. Adisak Muakdang

-Research Assistant , Department of Pharmacognosy and Pharmaceutical Botany.

              Name/Quantity of Solvent : DMSO

Type of container : 2 ML vial HPLC

              Temperature setting : without heating

              Operation hour : 9 hours

Q:What is your research ?
A:Ms. Similan : Analysis of andrographolide in Andrographis extraction ( or in thailand called Fa thalai chon)

Mr. Adisak : Analysis of Mitragynine and 7-hydroxy-mitragynine in Kratom leaf extract.  Mostly, we research about Local herb extraction. We analyze the active ingredient in the Local herb.

Q:Could you please tell us a little bit about your process?
A:Ms. Silmilan : I extract the Andrographis by using reflux extraction with 85% ethanol.

After Reflux Extraction, I use Smart Evaporator C10 to evaporate out the solvent.  Then I use HPLC to analyze the quantitative of andrographolide with water/acetonitrile as a mobile phase.

Q:That’s reason why you have to evaporate out the ethanol.
A:Ms. Silmilan : Yes, we use a different solvent so we have to evaporate all of the ethanol before HPLC.

Q:How about you Mr. Adisak ?
A:Mr. Adisak : In order to extract 7-hydroxy-mitragynine, we can not be use the chloroform

because 7-hydroxy-mitragynine can be decomposed in chloroform. We have to use DMSO as a solvent instead.  In order to evaporate out the DMSO, we have to control the temperature to not over 50°C.

Q:What is your method before using Smart Evaporator C10 ?
A:Mr. Adisak : Before using the Smart Evaporator C10, I used to work with Freeze dry machines before.

Ms. Similan : Previously using Rotary Evaporator, it took more time. (Takes about 30-40 minutes) Compared to using the Smart Evaporator C10 it takes only 10-15 minutes and can do 10 samples at once.

Q:What is your benefit you get from using Smart Evaporator C10 ?
A:Ms. Silmilan : We finish our workflow faster! We are concerned about the timing for our sample because the active

ingredient will decompose, if the laboratory took a long time. We have to complete the laboratory within one day. If we use Rotary Evaporator, we can do one by one sample and it would take a long time.  When we use Smart Evaporator C10, we can do 10 samples in time so it can be completed in a day.

Mr. Adisak : Compared to the use of Freeze dry, it can save the cost of using nitrogen gas.  It took me 9 hours, but I was able to do other things in the meantime.
